More woes - cylinder 5 misfire.

An interesting second day of ownership!

After getting home earlier I plugged the car into a fairly basic diagnostic reader I have.
As ever, on the initial scan, it found lots of codes but, I always find that many of these can be historic and not relevant/current so, I recorded them, cleared them and then restarted the car end went through the process again.

On the second diagnostic run it came up with the following issues;

P0300 misfire detection (total) exceeds limit value.
P0305 misfire, cylinder 5 exceeds limit value.
P1654 coolant shutoff valve exceeds limit value.

I'm going to do a little bit of research on the coolant code and work out what that could be but, I'm hoping the misfire could be as simple as a plug/coil pack but, I just thought I'd enquire here - I've had a brief look here but can't find anything obvious re cylinder 5 but, I was just wondering if there were any 'known' issues with cylinder 5 - near to something that can leak into it, proximity of a component that can cause issues, etc?

Apologies, I should have put a little bit more information!
It's a tiptronic and it drove fine (for 20 miles) and then the temperature failure warning light came on followed a few minutes later by the engine check light.
When this happened, the engine suddenly felt rough and it does feel like a misfire.
After that, the engine check light has varied from flashing to constant and the temperature failure light has 'rectified' itself temporarily and gone out and the gauge has functioned correctly, before failing again.
I can't find an obvious correlation between to two issues.
 
I'm guessing the misfire and cooling codes are 2 separate issues.

I'd:

1. Swap coil packs from left to right & vice versa.
2. The P1654 code on the tiptronic (after a quick root round on here and the web) could be pointing at an issue with the change over valve for the tiptronic cooling. It's on the side of the gearbox. It's an electronically operated vacuum switch.
